Glioblastoma remains a formidable brain cancer, with current treatments offering limited success due to the protective blood-brain barrier (BBB). New preclinical research has introduced a promising approach using a modified vitamin B12 analogue, nitrosylcobalamin (NO-Cbl). This compound was engineered to carry nitric oxide, a cell-killing substance, and leverages the high presence of CD320 receptors on cancer cells for uptake.
A significant finding is NO-Cbl's ability to penetrate the BBB in animal models, a major hurdle for brain cancer drug delivery. In rats with glioblastoma, NO-Cbl accumulated in tumor cells, with elevated nitric oxide markers persisting longer in cancerous tissue than in healthy cells. This targeted delivery system shows potential to increase treatment efficacy and reduce systemic toxicity.
Furthermore, NO-Cbl demonstrated synergistic effects when tested alongside temozolomide, a standard chemotherapy drug, and TRAIL, another anticancer agent. This combined approach proved more effective in suppressing tumors than either treatment alone in laboratory settings. While still in the preclinical phase, this research marks a crucial proof of concept for advanced drug delivery systems targeting challenging neurological cancers.
