Pancreatic Cancer Drug Shows Lung Cancer Promise

A breakthrough medication, daraxonrasib, initially approved for advanced pancreatic cancer, is now showing potential as a treatment for a specific type of lung cancer. In an early-phase clinical trial involving 136 patients with non-small cell lung cancer, daraxonrasib was administered daily. These patients had previously undergone other treatments without significant success and possessed RAS protein mutations, which are known to fuel tumor growth.

The study, published in the New England Journal of Medicine, found the drug to be safe and effective in reducing tumor burden in over 30% of participants. Doses of daraxonrasib, marketed as Rasonque, resulted in tumor reduction across different dosage groups, with rates ranging from 31% to 37%.

Despite its promise, the drug was associated with side effects in nearly all participants, with 54% experiencing severe reactions. Common adverse effects included rash, diarrhea, nausea, and vomiting, alongside mucositis, stomatitis, pneumonia, and low blood counts. The Food and Drug Administration (FDA) will likely require further large-scale trials before considering approval for lung cancer treatment.

This development offers potential hope for lung cancer, a leading cause of cancer-related deaths globally and in the U.S. The FDA had previously granted daraxonrasib a "Breakthrough Therapy" designation for pancreatic cancer, leading to expedited review and approval. A late-stage trial for pancreatic cancer showed that median overall survival nearly doubled for patients treated with daraxonrasib compared to standard chemotherapy.
