New Study: Tirzepatide Causes More Lean Mass Loss
17 Apr
Summary
- Tirzepatide shows greater weight loss but higher lean mass loss.
- Lean body mass loss with tirzepatide increases over 12 months.
- Decreased exercise tolerance linked to greater lean mass loss.

New research indicates that tirzepatide, an Eli Lilly drug, promotes more substantial weight loss than semaglutide, a Novo Nordisk medication. However, this increased weight reduction is accompanied by a greater loss of lean body mass, including muscle and connective tissues, according to findings published ahead of peer review.
The analysis of patient data revealed that tirzepatide users experienced a 1.1% greater lean body mass loss after three months and 2% more after 12 months compared to semaglutide users. This lean mass reduction was more pronounced in patients who experienced decreased exercise tolerance or had pre-existing musculoskeletal pain.
While both drugs mimic hormones to reduce appetite and slow digestion, the study could not definitively explain why tirzepatide resulted in more lean mass loss. Representatives from Novo Nordisk and Eli Lilly noted that lean body mass loss is typical with weight loss and did not differ significantly from placebo groups in their own clinical trials.
