J&J's Rybrevant Offers New Hope in Lung Cancer
14 Sep
Summary
- Rybrevant combination therapy achieved longest median survival.
- Advanced NSCLC patients lived nearly three years.
- New treatment significantly improves survival rates.
Johnson & Johnson's Rybrevant (amivantamab-vmjw) in combination with chemotherapy has shown unprecedented results in treating advanced non-small cell lung cancer (NSCLC) with EGFR exon 20 insertion (Ex20ins) mutations. The Phase III PAPILLON study reported a median overall survival (OS) of 34.3 months for patients receiving the bispecific antibody combination, marking the longest survival seen in this patient group to date.
These findings represent a significant advancement, as EGFR Ex20ins mutations have historically presented challenges for targeted therapies. The combination not only extended median OS by over six months compared to chemotherapy alone but also demonstrated a 43% reduction in the risk of death when accounting for crossover treatments. The safety profile remained consistent with previous studies, with common side effects including paronychia, neutropenia, and rash. These results, presented at the IASLC 2026 World Conference on Lung Cancer in Seoul, build upon earlier data showing a significant improvement in progression-free survival.
The historically low median OS for this condition, ranging from 16 to 24 months, underscores the importance of these new findings. Rybrevant is now approved for patients with common and Ex20ins EGFR-mutated advanced NSCLC in first- and second-line settings, offering a crucial new option in the evolving NSCLC therapeutic landscape.
